Marker-Assisted Pyramiding of Multiple Disease Resistance Genes in Coffee Genotypes (Coffea arabica)

The use of resistant cultivars is the most effective strategy for controlling coffee leaf rust caused by fungus Hemileia vastatrix. To assist development such cultivars, amplified fragment-length polymorphism (AFLP) markers linked to two loci resistance races I and II as well pathotype 001 H. vastatrix were converted sequence-characterized region (SCAR) cleaved polymorphic site (CAPS) markers. In total, 2 SCAR 1 CAPS marker validated in susceptible parents 247 individuals from F2 population. efficiency these marker-assisted selection (MAS) was evaluated F2:3 backcross (BCrs2) populations genotyped with developed phenotyped race showed 90% MAS. Therefore, markers, together molecular associated other genes, used F3:4 BCrs3 selection. selected plants analyzed using berry disease (CBD) resistance, aiming preventive breeding. MAS all feasible.
